Human serum albumin CY3 (HSA-CY3) is a widely used fluorescent probe in biochemical and biophysical research. The labeling process involves several key steps:

1. Solution Preparation: Dissolve HSA in a suitable buffer, such as 0.1 M sodium bicarbonate buffer (pH 8.3), at a concentration of 1-10 mg/mL. Dissolve CY3 NHS ester in a small volume of dimethyl sulfoxide (DMSO) to make a concentrated stock solution (1-10 mg/mL).

2. Labeling Reaction: Add the CY3 NHS ester solution to the HSA solution at a molar ratio of 5-10 moles of CY3 per mole of HSA. Incubate the mixture at room temperature for 1-2 hours with gentle stirring. The NHS ester reacts with primary amines on HSA, forming stable amide bonds.
