The DeffMACRO values of Cy3-DOPE were significantly increased and decreased by 30%, after the cells were treated with 5 µm latrunculin-B and 25 µm jasplakinolide, respectively (Figure 4, bottom right; Table 1), consistent with our previous observations in other cell types 19, 45, 48. These drug concentrations were much higher than those used previously, but PtK2 cells were extremely resistant to these drugs, and did not exhibit any morphological changes even under these conditions. Considering these results, along with the result showing a 20-fold increase of the Cy3-DOPE DeffMACRO value upon blebbing + actin depletion (Table 1), we concluded that the actin filaments associated with the PM cytoplasmic surface are responsible for slowing the diffusion of Cy3-DOPE in the PtK2 PM, as found previously in other cell types
